Well it depends on the competition you are referencing actually, so if you are talking about the Euro's then no South American World Cup qualifiers are more difficult because I think in the current format the top two from each group make it to the Euros with the rest making it through the playoffs.
However, European World Cup qualifying may actually be more difficult because only group winners qualify automatically while second and some third-place teams get put into the playoffs, and of the 12 teams that were in playoffs only three make it to the World Cup.
This means that overall, only 13 of 55 possible teams qualify for the World Cup which is 23 percent of the region. Compared to South America there are at least 4 of 10 teams are guaranteed a spot in the tournament which 40 percent and on some years, they have 5 teams as the 5th place team qualifies through the playoffs making half of the region making it in. So, from a numbers point of view, it would be harder to qualify in Europe than in South America.
Even if the argument is that they play weaker teams in Europe which, could be true that actually doesn't really benefit them in the long run because there a smaller margin for error in European qualifiers because the best teams in each group will win most of their matches meaning one draw could prevent be the difference between winning the group or possibly having to qualify through the playoff.
With South American World Cup qualifying there is a much greater margin for error meaning that teams do not need to be great or even good through most of qualifying and they still have a good chance qualifying for the World Cup Automatically or at worst making to a playoff where they play usually an inferior team from either Asia or New Zeeland.
It not uncommon to see teams talented in that region such as Argentina, Brazil, Columbia, and etc be awful for most of qualifying and still get in by picking up some form at the back end of the campaign to get into the competition. In European qualifying this wouldn't be the case as the margin for error of too slim for this happen in this region which is why it is probably harder to qualify from.
